Nadelen traditionele PCR Resultaten zijn niet erg nauwkeurig. Het duurt relatief lang voor de micro-organismen geïdentificeerd zijn. De opbrengst van de PCR varieert van monster tot monster. Deze techniek is hooguit semi-kwantitatief, er kunnen geen exacte hoeveelheden micro-organismen bepaald worden. Principe real-time De bindingsreactie kan zichtbaar gemaakt worden door het nagemaakte stukje DNA te voorzien van een fluorescerend label met quencher (een stofje dat de fluorescentie uitschakeld) Wanneer een organisme waarnaar gezocht wordt in het materiaal aanwezig is, zal dit gevlagde stukje DNA binden aan het specifieke DNA en later in het proces uit elkaar vallen, waarbij de quencher los komt van het fluorescerende label, zodat er licht uitgestraald kan worden. 16 RAC scholingsmiddag 29 september 2014
