Fysiologische rol koolzuur anhydrase ph < 7 ph ~ 7.4 [Figuur 7.22] Koolstofdioxide wordt in de rode bloedcel gehydrateerd. Het gevormde bicarbonaat wordt uitgewisseld voor chloride en zo via het bloed naar de longen vervoerd. Het CO 2 en de verlaging van de ph stimuleren de afgifte van zuurstof van hemoglobine tot 90% van zijn maximale transportcapaciteit.
Koolzuuranhydrase is een van de snelste enzymen. Elk enzymmolecuul kan per seconde 10 6 CO 2 moleculen per seconde hydrateren. De katalytische cyclus wordt in een-miljoenste seconde doorlopen.
Metalen zoals Zn en Ni zijn altijd stevig gebonden. Enzymen kunnen energie in een andere vorm omzetten Fotosynthese: licht(energie) in een chemische binding (NADPH en ATP). Uiteindelijk in gereduceerde koolstofverbindingen (glucose, vetzuren). In mitochondriën wordt energie aanwezig in voedsel via redoxreacties omgezet in een ionengradiënt die wordt gebruikt om ATP uit ADP en fosfaat te vormen. De bruikbare vrije energie die in ATP aanwezig is, wordt door enzymen gebruikt voor de contactie van spieren, voor actief transport van moleculen of voor de synthese van moleculen.

Enzymen versnellen reacties door de vorming van de overgangstoestand te bevorderen (8.3). v = factor x [s] x e -ΔG ǂ/RT RT = 2.47 kj.mol -1 e -5.71/2.47 = e -2.312 = 0.1 Het vrije energie verschil tussen de producten en dat van de substraten bepaalt de richting van de reactie (ΔG). Het vrije energieniveau van de overgangstoestand bepaalt de snelheid van de reactie (ΔG ).
Wanneer de overgangstoestand met 5.71 kj.mol -1 wordt gestabiliseerd, wordt de snelheid vertienvoudigd. Naast het stabiliseren van de overgangstoestand kan ook de vorming van het enzym-substraatcomplex een specifieke reactie mogelijk maken. De extra bindingsenergie die bij een specifieke binding vrijkomt wordt gebruikt om de DNA dubbelhelix te buigen waardoor water de fosfodiesterbinding kan bereiken en hydrolyseren. Figuur 9.38 en 9.42
