1 Curriculum vitae De auteur van dit proefschrift werd geboren op 5 november 1973 te Nijmegen. Zij behaalde het HAVO diploma in 1991 aan de Nijmeegse Scholengemeenschap Groenewoud te Nijmegen. In 1993 startte zij met de hogere laboratorium opleiding aan de Hogeschool van Arnhem en Nijmegen en vervolgde deze aan de Leidse Hogeschool te Leiderdorp. Tijdens deze studie heeft zij onderzoek verricht bij de afdeling Pathologie van het VU Medisch Centrum (VUmc) onder begeleiding van dr. Danny Dukers en dr. Lies Jaspars. In 1997 behaalde zij het HLO-diploma: afstudeerrichting cyto-histopathologie en werd aansluitend begonnen met de opleiding medische biologie aan de Vrije Universiteit te Amsterdam. In het kader van deze studie liep zij onderzoeksstage op de afdeling Moleculaire Genetica van het Nederlands Kanker Instituut te Amsterdam onder begeleiding van dr. Roderick Beijersbergen. Het doctoraal diploma, met als specialisatie Oncologie werd in 2000 behaald. Vervolgens begon zij aan haar promotie onderzoek binnen de afdeling Pathologie van het VUmc onder leiding van dr. Joost Oudejans en prof.dr. Chris Meijer. Dit en de resultaten ervan zijn in dit proefschrift weergegeven. Na haar promotie blijft zij vooralsnog werkzaam als onderzoeker op de afdeling Pathologie van het VUmc. The author of this thesis was born on November the 5th 1973 in Nijmegen, the Netherlands. She finished secondary education in 1991 at the Nijmeegse Scholengemeenschap Groenewoud in Nijmegen. In 1993 she started her BSc training at the Hogeschool of Arnhem/Nijmegen and continued this study at the Leidse Hogeschool in Leiderdorp. During this study she performed a scientific training at the department of Pathology at the VU University Medical Center (VUmc) under supervision of dr. Danny Dukers and dr. Lies Jaspars. In 1997 she completed this study and started her MSc training at the VU University in Amsterdam. She performed research at the department of Molecular Genetics at the Dutch Cancer Institute under supervision of dr. Roderick Beijersbergen. After her graduation in 2000 with the specialization on Oncology, she started working as a PhD student at the department of Pathology at the VUmc under supervision of dr. Joost Oudejans and prof.dr. Chris Meijer. The scientific research and results related to this subject are described in this thesis. As yet she is intending to continue her work as a post-doc at the department of Pathology of the VUmc. 126

5 Dankwoord En dan de meest gelezen en laatste pagina s van dit proefschrift, waarmee ik deze promotieperiode afrond. Een periode waar ik met veel plezier op terugkijk met pieken en dalen, maar waarin ik vooral veel geleerd heb en veel verschillende mensen heb ontmoet en mee heb samengewerkt. Dit is dan het moment om iedereen (et al.) te bedanken, die heeft meegeholpen, meegedacht, aangemoedigd, ondersteund of op een andere wijze heeft bijgedragen aan het gereedkomen van dit proefschrift. (ik hoop dat ik niemand vergeten ben) Allereerst mijn promotor, prof.dr. CJLM Meijer. Beste Chris, jouw uitgebreide kennis en enthousiasme over onderzoek in het algemeen en lymfomen in het bijzonder werkt aanstekelijk. 6 De afdeling Hematologie, die me vanaf het begin van mijn Aio-schap heeft geadopteerd. Bedankt voor het altijd even tussendoor kunnen FACSen, de hulp als de FACS niet deed wat ik wilde, de fijne donderdagmorgen discussies, de gezellige borrels en de prettige samenwerking! Corine, Fedor en Gerrit-Jan, ik heb veel van jullie geleerd over de RT-MLPA techniek en het analyseren van de data. Corine, bedankt voor de enthousiaste en gezellige gesprekken, succes met de laatste loodjes! Sander, lymfoomkoerier, bedankt voor het ophalen van lymfomen uit Zaandam en het sorten van samples. 7 Josine en Tabitha. Onze wandeltochten zijn altijd erg leuk en heerlijke momenten van ontspanning, hoewel. (caravans, zwanen en honden). Josine, ik kijk uit naar onze volgende trekvogeltocht met bijbehorend het te testen koffietentje. Tabitha, bedankt voor al de celkweek-sessies, telefoontjes, DE-momenten waarbij je een luisterend oor was. Ik vind het ontzettend leuk dat je bij mijn promotie aanwezig bent en ik wens je nog een mooie tijd in Schotland! Remco en Bart & co. Het was heerlijk om met jullie stoom af te blazen in ons stamcafé De Toog (soms iets later, dan de bedoeling was!). De avondjes met zinnige en onzinnige gesprekken vliegen altijd om. Over een half jaar in Zuid-Afrika of met Ukkie in Woerden? De mededirecteuren Maarten en Marcel. Onze enthousiaste vergaderingen vol humor en met notulen(!) in het DE-café waren een welkome afwisseling tijdens stressvolle tijden. Ik ben benieuwd waar ons avontuur ons brengt na de sabbatical. Marcel, veel succes met het afronden van jouw promotie, je bent er bijna! Nancy en Alex, Marjolein (Klavertje) en Paul. Nu hebben we eindelijk meer tijd om leuke dingen te gaan doen (dat is niet alleen zwemmen, Nance!). Ondanks de drukte (verhuizingen, proefschriften) hebben we heel wat afgelachen. Dank voor jullie steun en vriendschap! Het Vormburo (Niels!). Ik ben heel erg blij met de mooie lay-out en voorkant van het boekje. Heel erg bedankt dat je het in zo n korte tijd voor elkaar hebt gekregen, echt top! Tot slot de familie en vrienden. Jullie zijn heel belangrijk geweest tijdens deze promotieperiode. Ook al was het voor jullie soms moeilijk te begrijpen wat voor een onderzoek ik precies deed, jullie hebben altijd interesse getoond. Op de juiste momenten zorgden jullie voor de broodnodige ontspanning en gezelligheid. Bedankt voor het skiën, après-skiën, squashen, après-squashen, de saunabezoeken, zwemmen, te laat zijn, maar toch op tijd zijn, samen fluiten, de ensembledagen, DE-tjes, theetjes, BBQ s, (te) lange wandeltochten, fietstochten langs de Linge, high-tea-en, boerengolf, strandwandelingen, de 1890, de restaurantjes-testen-tocht, concerten in Ahoy, gezellig eten en drinken, etc! Lieve Jan-Michiel en Yvonne, jullie hebben altijd een warme belangstelling gehad voor mijn promotieonderzoek en allerlei goede adviezen gegeven. John-Dave, the plan of Zuster has come together! Yvonne, welke musical gaat het worden? Bedankt dat jullie er altijd zijn als het nodig is (ook na een Busstop!). Lieve pa en ma, zonder jullie onvoorwaardelijke steun was dit nooit gelukt! The results following unsupervised cluster analysis are shown. Two groups of DLBCL could be identified: one with relatively low levels of both pro- and anti-apoptotic gene expression, and one with high levels of apoptosisregulating gene expression. The expression levels of BCL-G, Harakiri, ciap, and BIRC7 were too low or not detectable. Cluster analysis was restricted to those genes that were expressed in at least 80% of the samples. Omitted genes were BNIP3 and Bik. (B) RT-MLPA analysis of apoptosis-regulating gene expression in cell lines. Unsupervised cluster analysis was performed. The intrinsic apoptosis pathway is constitutively activated in a subset of chemotherapy DLBCL with downstream disruption of this pathway. Previous RT-MLPA analysis revealed two groups of DLBCL: one with relatively low expression levels of both pro- and antiapoptotic genes (low apoptosis profile) and one group of DLBCL with relatively high expression levels of pro-apoptotic genes in combination with high expression levels of anti-apoptotic genes (high apoptosis profile). DLBCL with a high apoptosis profile frequently appeared to be chemotherapy-refractory and were characterized by spontaneous caspase 9 activation without induction of caspase 3 activation or apoptosis (A). Chemotherapy failed to induce apoptosis in DLBCL with a high apoptosis profile, due to inhibition of caspase 3 by XIAP protein expression (B). Low apoptosis profile cells showed no spontaneous caspase activation or apoptosis, only after induction with chemotherapy, induction of apoptosis was observed.
