Het betreft de vakgebieden farmacie, rechten, linguïstiek en geschiedenis & archeologie. De ranglijst is gebaseerd op wetenschappelijke reputatie, citaties en werkgeversimago. Natuurwetenschappelijk Gezelschap nieuw leven ingeblazen Tijdens een speciale bijeenkomst op donderdag 20 maart 2014, onthulde het Natuurwetenschappelijk Gezelschap Leiden (NGL), voorheen het Natuurkundig Gezelschap Leiden, samen met de decaan haar nieuwe koers, een mijlpaal in hun 144-jarige bestaan. Eind 2013 heeft het NGL in samenwerking met de Faculteit W&N om een nieuwe koers te varen en haar zichtbaarheid te vergroten. De nieuwe doelstellingen zijn terug te vinden op de nieuwe website. Twee Vici s voor Leidse wetenschappers Natuurkundige John van Noort en taalkundige Marian Klamer hebben van NWO een Vici toegekend gekregen. Met deze subsidie van anderhalf miljoen kunnen ze een eigen onderzoeksgroep opzetten en promovendi aanstellen. Scheikundige Huib Ovaa en farmacoloog Jos Jonkers (werkzaam bij het NKI) hebben ook een Vici ontvangen. Zij hebben ieder ook een kleine aanstelling in Leiden. Leiden krijgt unieke internationale biotechtrainingsfaciliteit Op het Bio Science Park is op 24 februari het contract getekend voor de bouw van een unieke Biotech-trainingsfaciliteit voor werknemers in de (bio-)farmaceutische industrie. Wereldwijd bestaan er slechts vijf tot tien vergelijkbare faciliteiten. De Biotech-trainingsfaciliteit is voor de Universiteit Leiden, de stad en de sector een belangrijke aanwinst. Groeien en uitvinden In Nederland is studeren voor de meesten iets vanzelfsprekends. Sterker nog: studenten krijgen geld, tijd en ruimte en zien dit als hun recht. Het wetenschappelijk klimaat van Nederland is goed en er zijn kansen voor uitvinden en ontwikkelen. Maar is dit nou eigenlijk wel echt iets vanzelfsprekends? Hoe gaat het er in andere landen er aan toe? Hoe ontwikkelen landen als Zuid-Afrika of China zich (pagina 4)? En wat heeft wetenschap deze landen te bieden (pagina 12)? Hoe zien scholen en steden er daar uit (pagina 26)? Daarnaast zijn er veel studenten die uit andere landen Nederland aandoen om kennis te vergaren, en genoeg Nederlanders die graag willen studeren in het buitenland. Hoe vergaat het deze mensen? In het studentenartikel komen beide kanten aan bod (pagina 8). Studeren is in andere landen wellicht ingewikkelder dan in Nederland. We mogen dan ook blij zijn met de prachtige faciliteiten en cultuur die we rijk zijn. Maar andere landen zijn ook niet verloren: wetenschap zorgt voor snelle ontwikkelingen, en het omgekeerde is misschien ook waar! Trouwens, ook ons blad ondervindt ontwikkelingen. But so are Brazil and South Korea, and the question that rises is: how are these countries changing themselves? Countries that are experiencing rapid growth are called tiger cubs. This special is all about the transition from a poor country to tiger cub and beyond. Also, we will take a closer look at some of these tiger cubs. The theory behind tiger cubs After the Middle Ages, Europe experienced rapid growth. For instance, Holland had the so-called Golden Age, in which trade, science, military, and art provided them with a lot of wealth and riches. Later on, the first fully industrialized region was West- Europe. North-America soon followed, but other continents lagged behind. This was partly a consequence of the period of colonisation many countries in Africa, South America and Asia were recovering from. The European countries that had power over the region during that period, used these countries only for their manpower and sources. Nothing was done to develop the government or economy in these countries. When the settlers finally left, the countries were ready to develop themselves. Unfortunately, this is quite a task to accomplish. Countries such as China had difficulties with copying the success industrialisation brought Europe. Some of them remained poor and undeveloped despite their efforts. Several things were to blame: Countries with a very closed economy would never get the tips and tricks about industrializing from more developed countries, which turned out to be a big disadvantage. Besides that, countries without a stable political situation also experienced trouble developing, as civil unrest and bad decisions held them back. A lot of ex-colonies lacked stability, because their former bosses didn t help them form a proper government. Before any progress could occur, this issue had to be fixed. Another name for the tiger phenomenon is NIC, which stands for Newly Industrialized Country. Industrialisation is therefore considered the base of the expanding growth a country experiences. One of the first tigers was Japan. After the Second World War, the United States aided in the reconstruction of Japan. The United States stimulated the industrial sections and tried to help economical planning. Beforehand, most of Japan s people worked in the industrial sector, but after this aid from the USA, wealth increased and Japan s own manpower became too expensive. A lot of industrial activity was therefore moved to countries where people would work for lower wages. These low-wage countries benefited from this movement, but this could not have happened if they didn t have the business climate they had, created by their strong governments. These low-wage countries became great growers after Japan pushed their economy and industry. These low-wage countries became the first generation tigers. They were the first to expand so rapidly without direct donations from outside. By growing so swiftly, the wages for simple industrial work in the tiger cubs also became too high. This is where a new generation is needed: a new group of countries where the business climate is attractive and lots of cheap manpower is available. With this mechanism, the second and third generation tigers were born. These countries are the new 21 st century tiger cubs. Most well-known NIC s are found in Asia. A few examples are (first, second and third): Singapore, Malaysia, and China. But there are a few tigers in other continents as well. Imagine South Africa and another very famous tiger: Brazil. These countries often have interesting backgrounds, so it is time to look a little closer at some of them. The first generation giant A first generation tiger cub with a unique story is Singapore. Earlier, I mentioned the importance of a country s government, and this little state in Asia is a great example. Singapore was one of the first generation tiger cubs, and the most prosperous South-East Asian country. The country already had a strong economy and could be called a tiger cub beforehand, but with the arrival of multinationals from all over the world, it surely became a full-fledged tiger. Singapore had soon become a hub for a variety of product transportations. These people organised themselves in science parks and invested in research and development In 1980, the wages became too high for the industry in their own country, and they outsourced labour to the second generation of tigers. Singapore decided to develop a more knowledge based industry, for example the pharmaceutical industry. Through the last years of development, Singapore gained a group of highly educated people, and these people stayed (a lot of developing countries experience brain drain: a phenomenon where the highly educated group emigrate). These people organised themselves in science parks and invested in research and development. This strategy worked great and these workplaces became very rich. Singapore is now a country capable of competing on both a regional and global scale.

6 6 ORIGIN # 3 jaargang 9, april 2014 So how did Singapore become such a successful country? The answer lies in the high amount of political control that is and was exerted. Singapore is famous for having a more authoritarian government, rather than democratic. But, unlike in some other authoritarian governments, corruption didn t kill business, and a very transparent economy was formed. Through the strict performance, the business climate was shaped successfully and a lot of bonds with neighbouring and overseas countries were made. This turned out to be a good foundation for a successful Singapore. A second generation star One of the countries that started growing thanks to outsourced projects from Singapore is Malaysia. This neighbouring country belongs to Asia s second generation tiger cubs. In Malaysia, the first period of growth was thanks to the low wages of its workforce. Another advantage Malaysia had were the many natural resources. The export of these products were still a important part of the economy. Unfortunately, for complicated economical reasons these projects became less valuable in the 1980 s. Malaysia chose another path, called import substitution, which means making products you otherwise have to import. This can save a lot of money and creates employment while you re at it. After establishing this, Malaysia focused more on the export related industries, and this transition caused a further increase in wealth. Around the end of the 1990 s, the labour-intensive projects were again moved to lower-wage countries: the third generation tigers. Unlike Singapore, the key to success for Malaysia was the switch to import substitution and export related industries. Why was this important? Import substitution has a lot of advantages for growing countries when they can pull it off. Instead of buying products from other countries, which makes you dependent of them, you only have your own production costs, and you can Import substitution has a lot of advantages for growing countries when they can pull it off sell these products with profit for yourself. The production itself creates a lot of work for the poorer part of the society. By using this strategy, the entire country can grow, instead of only the richer merchants or politicians. The other part of Malaysia s strategy, the export related industries, were beneficial because this plan was very attractive for its rich neighbours. The rich countries could invest in products produced abroad, and shared in the profits. A lot of multinationals show interest in these kind of undertakings. What a lot of multinationals

7 DOSSIER OPKOMST Universiteit Leiden 7 do, is create a good working place, and improve the cities in the vicinity. It seems obvious why this is good for the economy of such a country: better roads for transport, for instance, improves the entire infrastructure. So in the end, not only the multinationals profit; the entire country does. But what about China? Where after the world wars, Japan quickly buckled up for a Western race to wealth, China was on a slightly bumpier road. First it had a very conservative government, then a communist one famous for starving its population. However, China is recovering, and remains an enormous country with a population of billion people. A large part of this population remains poor. But for China, the sheer amount of inhabitants is actually an advantage. So many people willing to work grants this country a lot of cheap manpower. This made China perhaps the strongest tiger of them all. So many people willing to work grants this country very much cheap manpower The term shows the transition in economical power. The countries seen as these BRIC are the big growers, and the prediction is that in 2050 these countries are the richest of the world. One of the benefits of belonging to these lucky few is the trust other countries place in these promises, which can lead to more foreign investors and investments. The benefits that result give Brazil the opportunity to grow further, shaping a nice circle. To this list of growing countries we have been calling tiger cubs, many more countries have applied. After all, they are otherwise called the N-11 (Next Eleven), BRICM (Mexico included), BRICET (with Eastern Europe) and BRICS (including South-Africa). This confusion shows that, despite the recession, many countries are grasping opportunities. When that many countries are growing, is every country going to be rich? Perhaps they will, but they can t all be the richest. There are certainly countries that will lose their top 20 status, and chances are that these will be close to home. Only time will tell, as many theories exist. But some experts say that the western world has lost its flexibility, and that the little tiger cubs might just tear its current status to shreds. Whether this will happen or not, remains to be seen. But no matter what happens, it seems like the future is bright for the tigers. And perhaps even brighter for their cubs. China was first used as a low-wage option for Japanese and Korean manufacturers to make their cheap products. That was the time where all our toys were labeled Made in China. We are now seeing a change: China is climbing. China s people began inventing and producing more products by themselves, which led to a rapid growth of cities and the upperclass. There are still a lot of poor people left, however, so labour has remained cheap. Products and their parts are still often from China, so is China going to get rid of their low-wage country status or not? The answer depends on where you look. While a large upper-class is swiftly getting bigger, richer and more educated, a large part of the community is still struggling. With farmers still moving to cities to make a living, forcing people into factories is still an easy option. China is benefiting from both being very rich, and being very poor at the same time. The other side of the planet Known for its beautiful rain forests, Brazil has also another valuable quality: its growing economy. The last decade was a wonderful one for the country, as its economy grew staggeringly. They have expanded the trade with other countries, like the United States, Japan and the European Union. These trade bonds are mainly export connections. Brazil is first of the world exporter of for example sugar, coffee, airplane parts and oranges. An important part of the position of Brazil is the membership of being a BRIC-country. BRIC stands for Brazil, Russia, India, China, also referred to as the Big Four.

8 STUDENTEN 8 ORIGIN # 3 jaargang 9, april 2014 Life in The Netherlands Sinziana Cristea is a foreign student who came to Leiden for the research master Bio-pharmaceutical sciences. She talks about the study Pharmacy in Romania and sheds a light on how she has experienced life in the Netherlands. Let s begin the same way I ve started all my official writings since February 2013: I am a master student in Bio-Pharmaceutical Sciences, at Leiden University, specialising in what I was most fond of during Pharmacy School: Pharmacology. I took the decision of leaving my close friends and family back home in Romania, because I wanted to do something different than most people in my field. Sinziana Cristea Master student Bio-Pharmaceutical Sciences Normally, all pharmacy students have to study for five years about various kinds and interesting subjects like: Chemistry, Pharmacognosy, Physiology and Pathology, Clinical Pharmacology, Toxicology, Management and Marketing. For each of these subjects there is also practical work involved, sometimes more than twenty hours per week. Furthermore, during each summer holiday every student is required to do an internship in a community pharmacy. In the

9 DOSSIER OPKOMST Universiteit Leiden 9 fifth year, the last six months of your studies are dedicated to a final community pharmacy internship. Also, during this last year the students are required to work on research projects in a department of their own choice. In most cases, considering the several mandatory community pharmacy internships, you end up as a community pharmacist after graduating, running or working at a big pharmaceutical chain that is mostly focused on selling drugs. So, all the reasons that you had to study Pharmacy in the first place are slowly fading away in favour of new marketing strategies. Another career path would be to study further for another three years specialising in Clinical Pharmacology or Clinical Laboratory Analysis and become a Hospital Pharmacist. Unfortunately, there is still much space for improvement when it comes to the equipment used in Romanian hospitals. Micro-production machinery and research laboratories are currently available only in research institutes and university hospitals, outside the university setting. In this case, what you can do as a hospital pharmacist is restrained to filling in patients charts and giving them the right medication in the right dose, as prescribed by the doctor. Last but not least there is research. If you feel you have the skills for this, you decide to pursue a career in academia. If you are lucky, you get a PhD position at one of the departments with vacancies together with a nice scholarship. If not, it is also possible to get a PhD position and pay a tuition fee for it. As a PhD student, apart from your research project, you are also required to get involved in the education of the Pharmacy School students and help with preparing the practical work assignments and exams and also with supervising the usual practicals once or twice a week. Commonly, similar to the situation in The Netherlands, a PhD position has the length of four years with the possibility to apply for an extension of a maximum of two years. From what I am aware of at the moment, both the Faculty of Pharmacy of Bucharest and faculties from other universities throughout the country are involved in interesting research programs in collaboration with other European countries. There are some famous discoveries made by Romanians that I would like to mention: George Emil Palade studied mitochondria and established the role of these organelles within the cell. Also, he discovered the ribosomes (also called Palade s corpuscle), Nicolae Paulescu is one of the discoverers of insulin and, among more Romanian recent discovery are the telocytes or interstitial Cajallike cells that were discovered by the group of Professor Laurentiu M. Popescu in Bucharest. Unfortunately, even though representative research is still conducted in Romania, it is not really well known and funding for it is scarce. On the other hand, research here in the Netherlands has better funding, more collaborators worldwide and it is more popular as well. Furthermore, I think that the technical characteristics and equipment are a lot better. I must say that, at first, I was very impressed to see all of the research techniques being used at all the departments within the LACDR. Another thing that I find exciting and different is the Dutch lifestyle. The people seem to be relaxed and happy. The city planning is completely different from Eastern European cities and very well preserved. Going sightseeing is simply great, strolling through the parks you can see all these cute stray rabbits and all kinds of ducks and swans peacefully swimming on a lake or on a canal. Also, Dutch people seem to be exercising more than Romanian people do. Even in the cold season there are people jogging outside, biking, rowing, horse riding or doing all kinds of other sports activities that they enjoy. Another thing that I am glad about is the fact that people here are at ease with speaking English, which makes it easier for foreigners to feel integrated. I was very surprised that I could speak English even at the market and people would respond politely and patiently try to understand and help me as best as they could. On the other hand, in my experience so far, there are two things that I found really disappointing: the housing service for foreign students and the communal transport services for students. When I first arrived in the Netherlands, I found accommodation by myself in Amsterdam with some friends. After half a year of commuting I decided to apply for student housing in Leiden. This experience turned out to be fulfilling from a social point of view. I have made very nice and interesting international friends, but as for the living conditions Well, this was really problematic. The place resembled a very old refugee camp located somewhere outside the city, built on a former swamp, three kilometres away from the nearest supermarket. From the very beginning we had to deal with problems such as: slow to no functional internet connection; floods in the bathroom, common area and dormitories; rodents and no heating during the cold season. All of this occurred, even though the place was quite expensive. I must say that I would have never expected such poor living conditions in such a well developed country. To conclude, I believe that the overall experience I had so far was highly beneficial for me. I have met some very special people that made this experience enjoyable and unique and, as for research, now I think that I have a broader view upon what can be achieved within the field of Pharmacology.

10 10 ORIGIN # 3 jaargang 9, april 2014 Study Abroad: saving the severely threatened Philippine crocodile Protecting an animal from extinction is hard as it is. But, protecting an animal is even harder if the animal in question is associated with corrupt politicians and has a low cuddle-factor. Niels Fluitman Master student Bio-Pharmaceutical Sciences These things are the case for the Philippine crocodile (Crocodylesmindorensis): a severely threatened crocodilian which the Mabuwaya Foundation - in collaboration with Leiden University and the Isabela State University - tries to protect. As one of the many students lucky enough to get a taste of this partnership, I would like to share my experiences and tell something about the conservational features. The co-operation between these universities and a foundation seems odd at first, so let me explain a bit about its origin. The Mabuwaya foundation has arisen from the CVPED (Cagayan Valley Programme on Environment and Development); an organisation set up by Leiden University and Isabela State University. In the eighties the board of LU facilitated two partnerships with thirdworld-countries: one in Sub-Saharan Africa and one in South-East

11 DOSSIER OPKOMST Universiteit Leiden 11 Asia. Make a wild guess which country they picked for South-East Asia. Right: the Philippines. The main goal of this collaboration was enhancing international relations and easing the exchange of scientific information. For example, anthropological knowledge regarding the wide variety of cultures in this archipelago and biological information on the biodiversity of the islands. Consequently, the CVPED was formed. One of the many projects it established was regarding the conservation of the endemic Philippine Crocodile. The project was called: Croc-Project. The history of the Spanish colonization before the 19th century, followed by the colonization of the United-States and the dictatorial regime of Ferdinand Marcos, teaches that human influence has taken its toll on the natural habitat of the crocodilian. To put things into perspective: during the sixties, five ton of crocodile leather was exported annually. After word came out there was still a small population left in the wild, they decided to protect it. And a small population it is: only two hundred individuals remain in the wild. In context, the amount of lions (P. leo), also a threatened species, are estimated around individuals. After a few decades of successful conservation the Croc-project ended in 2003 and was incorporated into the independently operating Mabuwaya Foundation. What does Mabuwaya do to prevent the species from extinction? How do they operate? The term associated with this type of conservation is community-based. It implies a type of animal protection in co-operation with the local community. The people living in close proximity to the crocodile are involved in the protection and educated about the species. Besides, the organisation assisted in digging wells to prevent people from getting close to the crocodile-habitat, which are small creeks and water bodies. Assisting in land management is also one of the requirements for this kind of conservation. In highly impoverished areas the main crops are mostly fast growing and relatively low profit crops such as corn and cassava. Farmers in these kind of areas do not bother with long time investments such as fruiting trees. An external actor is necessary to carry the risk. The foundation does such a thing. Moreover, it assists local government in assigning fish sanctuaries. In these sanctuaries it is prohibited to fish, giving the aquatic species a safe haven for reproduction. Again, without an external actor, the local community would have never implemented such regulations. The methodology of the organisation also include active participation of the local community. For example, several, so-called BantaySanktuwaryo or sanctuary guards, have been assigned. These Filipino men are involved in the conservation and earn a modest incentive and medical insurance from the local government. They play an important role in informing the community and addressing threats. A recent study indicated that this type of conservation made the public positive about the crocodile and even proud of hosting such an iconic species. There is no place for pessimism regarding the conservation of the crocodile. In an age where iconic species such as the panda and tiger are strongly decreasing in numbers any effort regarding conservation is more than welcome.

12 12 ORIGIN # 3 jaargang 9, april 2014 Bètavraagbaak Opkomende een Derde Opkomende wetenschap is een fenomeen dat we zien in economische groeiers, stabiele marktaanvoerders en dus vooral in de landen waar het geld niet meer zo schaars is. Hoe komt een Derde Wereldland dan aan opkomende wetenschap en wat voor voordelen heeft dat voor zo n land? Piramide van Maslow Om te kunnen begrijpen hoe het komt dat een rijker en verder ontwikkeld land een grotere bijdrage levert aan de wetenschap dan een Derde Wereldland, pakken we het volgende figuur erbij: de Piramide van Maslow. Deze piramide laat de eerste levensbehoeften zien, met onderin de lichamelijke behoeften. Hieronder verstaan we alles dat mensen nodig hebben om hun lichaam goed te laten functioneren, zoals slaap en voedsel. Andere voorbeelden hiervan zijn de behoefte aan lichaamsbeweging en een algehele goede gezondheid. De volgende stap op de piramide is Rebecca van Rijn Bachelor student Biologie behoefte aan veiligheid en zekerheid. Mensen zoeken deze veiligheid bij een groep. Ook het hebben van werk, een vast inkomen en een betaalbaar dak boven je hoofd vallen hier onder. Als derde laag is er de behoefte aan sociaal contact. Daartoe behoren vanzelfsprekend vriendschap en (liefdes) relaties. Ook wat minder intense, maar wel positieve contacten, zoals het hebben van een goede buur vallen binnen de derde laag. Daarboven staat de behoefte aan waardering en erkenning. Dit wil zeggen dat mensen belang hechten aan sociale status en graag concurreren met anderen. Op het topje van de piramide staat de behoefte aan zelfontplooiing: het willen benutten van verschillende groeimogelijkheden. Belangrijk voor onze centrale vraag is dat leren een van de dingen is die we onder zelfontplooiing verstaan (en als gevolg daarvan het deelnemen aan de wetenschap). Maar wat heeft deze piramide nu te maken met wel, geen of weinig wetenschap in een land? Volgens de bedenker van deze piramide, Zelfontplooing Behoefte aan waardering en erkenning Behoefte aan sociaal contact Behoefte aan veiligheid en zekerheid Lichamelijke behoeften Abraham Maslow, moeten voor het beklimmen van de piramide eerst alle voorgaande lagen in orde zijn. Voorbeeld: om sociaal contact aan te gaan, moet je je eerst veilig voelen en moet er aan je lichamelijke behoeften zijn voldaan. In rijkere, verder ontwikkelde landen werkt deze piramide bij een groot deel van de bevolking als volgt: de lichamelijke behoeften zijn geregeld (genoeg eten en elke nacht lekker slapen), ook heb je de behoefte aan veiligheid en zekerheid vervuld door het dak boven je hoofd (dat je hebt betaald met je vaste baan). Met deze zekerheden als basis, kom je bij de behoefte aan sociaal contact. Deze behoefte wordt vervuld door collega s, een partner, vrienden of buren. Met de waardering en erkenning van je baas en collega s wordt ook aan deze behoefte voldaan, en dan is er ruimte voor zelfontplooiing. Deze zelfontplooiing kan vorm krijgen als creatieve ideeën op het werk, maar ook als een onderzoek dat iets oplevert voor de hele bevolking. Situatie in Derde Wereldlanden De situatie in veel Derde Wereldlanden is vaak heel anders: in de armste landen komt een deel van de bevolking niet eens aan het vervullen van de lichamelijke behoeften toe. En wanneer er sprake is van oorlog of sociale onrust, voelen inwoners zich ook niet veilig, waardoor ze enkel de tweede laag van de piramide bereiken. Door deze factoren, die natuurlijk weinig met wetenschap te

13 DOSSIER OPKOMST Universiteit Leiden 13 wetenschap in Wereldland? maken hebben, komen de mensen in Derde Wereldlanden niet toe aan zelfontplooiing. En geen ruimte voor zelfontplooiing betekent eigenlijk ook dat er geen ruimte is voor wetenschap. We zijn nu dan ook tegen het probleem van Derde Wereldlanden aangelopen: er is misschien wel vraag naar wetenschap, maar er is geen sprake van een milieu waarin dit kan worden uitgevoerd. Als er dus wel sprake is van opkomende wetenschap in een Derde Wereldland, wil dit zeggen dat het land al een aantal stappen in de goede richting heeft gezet. Er is tenminste bij een aantal mensen ruimte voor zelfontplooiing en dit heeft in het land geleid tot een groeiende wetenschappelijke sector. Hoe gaat dit voordelen opleveren voor mensen die niet bij de elite horen? Bijvoorbeeld door het verbeteren van de landbouw. In Derde Wereldlanden is vaak het grootste deel van de bevolking aan het werk om iedereen van voedsel te voorzien, zelfs al levert dit weinig geld op. Als de wetenschapssector daar aandacht aan besteedt, kan men wellicht de productie van voedsel efficiënter maken. Met efficiëntere landbouw zijn er minder mensen nodig voor de productie van dezelfde hoeveelheid voedsel, wat betekent dat een deel van de mensen ander werk kan gaan doen. Voorbeelden van deze groene uitvindingen zijn kunstmest, irrigatiesystemen en bestrijdingsmiddelen. Potentiële groei Kenmerkend voor ontwikkelende landen is dat niet langer de landbouw, maar de industriesector het hoogste percentage van de bevolking werk verschaft. De eerste productie zal vooral bestaan uit opdrachten die worden gegeven door andere landen. De echte opkomst van eigen industrie hangt meer samen met een opkomende derde sector: de commerciële sector. Een (vaak gebruikte) maatstaaf voor ontwikkeling is percentage bevolking werkend in de commerciële sector: hoe meer mensen er in de commerciële sector werken, hoe beter ontwikkeld een land is. Als de groei van de secundaire (industriële) en de tertiaire (commerciële) sector samenhangt, zal een opkomende wetenschapssector voor een sneeuwbal effect zorgen. De nieuwe ontdekkingen en visies die ontwikkeld worden door de wetenschap kunnen zorgen voor efficiënter werken en nieuwe investeringsmogelijkheden (sommige uitvindingen leiden tot nieuwe, vaak bijna automatisch. Een voorbeeld daarvan is de ontwikkeling van elektriciteit, wat snel leidde tot allerlei apparaten die daarvan gebruik konden maken, zoals de telefoon). Deze investeringen zorgen dan weer voor meer buitenlandse interesse. Met meer geld en investeringen wordt de mondiale positie van dit (voormalig Derde Wereld-) land verstevigd. Er zijn ook landen die deze theorie kunnen bevestigen : sinds de revolutie in Syrië (het verdwijnen van volledige tweede laag (de veiligheid) zijn overal in Syrië de educatiesystemen ingestort. Kinderen kunnen in hun eigen land niet meer naar school, omdat de toplaag met het verlies van de tweede is weggevallen. Toenemende wetenschap kan dus zorgen voor een plekje tussen de toppers van de wereld. Helaas zijn er wel wat voorwaarden aan verbonden voor een Derde Wereldland om daarvoor in aanmerking te komen: tenminste een deel van de bewoners moet in hun behoeften voorzien worden en streven naar zelfontplooiing (natuurlijk had de elite van zo n Derde Wereldland daar al tijd voor, maar er moeten meer mensen bij deze elite gaan horen voordat de elite iets kan bereiken voor het hele land). Met die instelling kunnen ze de condities in het hele land verbeteren en daarmee de sneeuwbal aan het rollen brengen. Dat wetenschap kenmerkend is voor rijke landen, komt alleen doordat zij al eerder ontdekt hebben wat je aan de top nodig hebt: innovatieve mensen, een goed ondernemersklimaat en buitenlandse investeringen. Ook deze landen hebben immers de transitie doorlopen van landbouwgrond naar kantorencomplex. Voor opkomende Derde Wereldlanden lijkt het dus enkel een kwestie van tijd.

In de eierenproductie kost dit wereldwijd miljoenen haantjes het leven, zij kunnen geen eieren leggen en zijn niet rendabel genoeg om te worden vetgemest tot slachtkip. De mannetjes zijn geen lang leven gegund en worden enkele ogenblikken nadat ze zijn uitgekomen vergast. In Nederland alleen al gaat het jaarlijks om 45 miljoen haantjes. Maar er is hoop, en een antwoord komt uit Leiden. Origin redacteur Rembrandt Donkersloot interviewt bioloog Wouter Bruins, die inmiddels al bekent staat als de haantjesredder. Hij heeft namelijk een oplossing voor handen Moeizame start Ik kom aan bij het Sylvius gebouw, waar ik als natuurkunde student niet vaak een bezoek aan heb hoeven te brengen. Ik kijk onwennig om me heen en probeer uit te zoeken waar de plek is waar ik heb afgesproken. Dan zie ik opeens Wouter met 2 koppen koffie aanlopen. Een betere ontvangst kon ik niet wensen! We gaan ergens zitten en ik vraag Wouter naar zijn achtergrond. Mijn studie begon in Leiden, vertelt Wouter, psychologie. Wouter vertelt dat dit geen goede keuze voor hem was geweest. Hij kon er zijn creativiteit niet in kwijt en de studie was niet concreet genoeg. Al snel kreeg Wouter genoeg van psychologie en begon aan een andere opleiding, economie aan de Vrije Universiteit van Amsterdam. Ook geen succes, aldus Wouter. Ondanks dat de eerste twee jaar van zijn studentenleven op het gebied van studiesucces niet geslaagd waren, ziet Wouter deze tijd niet als verloren jaren. In het bijzonder was Wouter positief over zijn lidmaatschap bij roeivereniging Njord, waar hij wijze levenslessen heeft geleerd en later roeicoach is geworden. Wouter wist niet goed welke richting hij wilde opgaan en besloot in plaats van een nieuwe studie te kiezen voor een jaar te gaan werken als koerier voor een apotheek. Mijn zus zei het al jarenlang, je moet biologie gaan studeren! Na een jaar gewerkt te hebben en alles op een rijtje te hebben staan kwam ik op dezelfde conclusie uit. Daarna ging het snel. Binnen tweeënhalf jaar wist Wouter zijn bachelor te halen. Zijn master heeft hij onlangs binnen gehaald, cum laude. Tijdens zijn tweede studie in Leiden heeft Wouter zich veel laten betrekken binnen de medezeggenschap, en heeft zo een groot netwerk opgebouwd. Dit kwam

15 DOSSIER OPKOMST Universiteit Leiden 15 later zeer goed van pas. Zo heeft Wouter niet alleen in de opleidingscommissie en faculteitsraad deelgenomen, maar heeft ook in 2009 een jaar zitting gehad in het faculteitsbestuur als student-lid. Student ondernemer Tijdens de masterfase koos Wouter om het Science Based Business (SBB) traject in te gaan. Eén van deze vakken, Entrepeneurship, heeft veel indruk op me gemaakt. Tijdens dit vak werden studenten erop uitgestuurd om ervaren ondernemers te interviewen. Wouter vertelt dat hij een aantal bedrijven in het Bio Science Park heeft bezocht. In een open sfeer leerde hij ervaren ondernemers kennen. Het waren mensen die met technologie het verschil wisten te maken. Het bezoek aan het Bio Science Park was dermate inspirerend dat het Wouter aanspoorde te gaan ondernemen. Het gevoel begon te komen om iets op te zetten. De vraag was nu alleen, wat? Het haantjes probleem Waar de meeste mensen hun inspiratie zochten op het internet, zocht Wouter zijn ideeën in de wijde wereld. Hij dook in een berg onbenutte patenten, op zoek naar een potentiele oplossing voor een probleem waarbij biologie een antwoord kan bieden. Zo kwam Wouter in contact met hoogleraren van het academisch ziekenhuis. Hier ontmoette hij ook zijn latere rechterhand, Wil Schuttenheim. Vanuit SBB werd de opdracht gegeven een businessplan te schrijven. De opdracht was om een (wetenschappelijke) oplossing te vinden voor een maatschappelijk probleem. De zoektocht was geopend. Wouter besefte dat je nu, als student, bij bijna elk bedrijf kan binnenlopen. Nu is het de truc om een groot problemen te zoeken dat speelt binnen een sector. Zo stuitte Wouter op het probleem met de haantjes na een gesprek met een kippenboer. Het merkwaardige was dat voor het onnodige dierenleed nog geen enkele oplossing bestond. Toen kwam de ingeving. Wat nu als we het geslacht van een kuiken al kunnen bepalen voordat het is uitgekomen? Al bekend was dat het kippengeslacht tussen dag 6 en dag 11 bekend was. Zou het geslacht nu ook al daadwerkelijk bepaald kunnen worden binnen deze termijn? Wouter ging samen met Wil met deze vraag aan de slag en ontdekte in het lab een biochemische methode die deze vraag daadwerkelijk zou kunnen beantwoorden. De universiteit bood Wouter en Wil de faciliteiten om de opdracht uit te werken, en ze deden dit met succes. Dit werd de basis voor het bedrijf In Ovo, dat Wouter en Wil samen op hebben gezet in Ze kunnen nu het geslacht vanaf de 9e dag al bepalen. Er is hiermee echter nog geen kuiken gered. Het is nu de opdracht om de methode op te schalen voor de industrie, en de meting per ei sneller te maken. Op die manier kunnen alle mannelijke eieren uit de productie worden gehaald en geütiliseerd worden. De kuikens zullen in deze fase er niets van merken en het onnodig leed zal zo bespaard blijven. The term Jugaad originates from Asia and means something like work-around. Especially in India they are very good at using this philosophy to find alternative ways to fulfill their needs. Good examples are the so called Jugaad motor vehicles, used as a cheap form of transportation. Their builders make use of scrap of motor cycles, tractors and sometimes even washing machines. A diesel engine, most often formerly used as irrigation pump, and some colorful tiling, make them a welcome addition to the Indian streets. Or aren t they? Bad breaks in combination with heavy loads have led to so many accidents that the Indian government has officially banned the Jugaad vehicles. 18 18 ORIGIN # 3 jaargang 9, april 2014 Institute Solving Cell Division Puzzles by Correlative Cryo-Light and Electron Tomography Streptomyces are Gram-positive multicellular soil-dwelling bacteria which are commercially used as natural producers of antibiotics, anticancer agents and immunosuppressants, as well as many industrial enzymes (Hopwood 2007). These microbes have an extraordinary ability to adapt to a diversity of ecosystems and thrive alongside a variety of competing and coexisting organisms. To do so, they have evolved a complex multicellular lifecycle, including several steps of morphological differentiation, with development tightly linked to antibiotic production (van Wezel and McDowall 2011; McCormick and Flärdh 2012). This life cycle starts with the germination of a spore, from which one or more germ tubes emerge. A complex mycelial network is formed as the hyphae grow and branch to form what is called the vegetative or substrate mycelium (Chater and Losick 1997). Growth occurs by tip extension, during which newly synthesized wall material is incorporated at the hyphal apex (Gray, Gooday et al. 1990). In Streptomyces, the combination of tip growth and branching results in exponential growth of the organism (Jakimowicz and van Wezel 2012). Nutrient depletion and other stresses (heat, drought, competing microbes) are considered the major triggers for the onset of aerial mycelium formation in the soil and also on solid-grown cultures in the laboratory environment. At this point, hydrophobic vegetative hyphae break through the moist surface of the soil or agar and grow into the air (Claessen, de Jong et al. 2006). Multiple cell division occurs, whereby up to 100 division septa are formed more or less simultaneously, and the multigenomic aerial hyphae are converted into chains of unigenomic spores. These spores then disseminate to start the formation of new colonies in more favourable conditions. In liquid-grown cultures, on the other hand, most streptomycetes stay in their vegetative state and form a branched mycelium, taking on a variety of morphological shapes, from dense pellets to small fragmented mycelia. In general, the morphology of liquid-culture pellets is a determining factor for productivity during industrial fermentations of filamentous organisms (van Wezel, McKenzie et al. 2009), with larger clumps favoring antibiotic production in Saccharopolyspora erythraea (Wardell, Stocks et al. 2002), and fragmentation stimulating enzyme production by Streptomyces lividans (van Wezel, Krabben et al. 2006). The understanding and control of mycelial morphology is thus key for optimization of industrial fermentations (Celler, Picioreanu et al. 2012). With this in mind, the purpose of my PhD project was to combine different approaches and techniques to arrive at a better understanding of Streptomyces morphogenesis and development, with the ultimate aim to improve Streptomyces as a production platform in industrial biotechnology. To this end, one of my major research lines involved using cryo-electron tomography (cryo-et) to study the ultrastructure of Streptomyces hyphae. Cryo-ET provides a unique opportunity to visualize and increase fundamental understanding of morphogenesis, capturing the ultrastructure of tip growth, branching, and cell division at nanometer resolution. Work on my thesis coincided with the opening of the Netherlands Centre for Electron Nanoscopy (NeCEN) in 2012, and so, in close collaboration with Dr. Roman Koning from the LUMC, I was able to use the new 300 kev Titan microscope to image vegetative cell division as no one has seen it before. In fact, in the past years, the possibility to image biological samples at high kev has been revolutionizing the field of bacterial cell biology and providing exciting new insights into macromolecular structure-function relationships in bacteria. 3D reconstructions, or tomograms, of tilt series images collected with the electron microscope provide an inside-view of cryo-fixed cells at conditions directly relevant to the native state of the cell, avoiding the artifacts of chemical fixatives. Building on this technology, we also developed cryo-correlative light and electron tomog-

Cryo-CLET correlates the images acquired by fluorescence light microscopy and electron tomography, enabling selective labeling and identification of cellular components as well as DNA and mapping of these lower resolution data onto high resolution tomograms. Investigating Streptomyces cell division in young vegetative samples, we found that large membrane structures form within hyphae prior to crosswall formation (see figure below). Combining cryo-clet and live fluorescent light microscopy, we discovered that the membranes, enriched in phosphatidylglyderol and cardiolipin, dynamically localize during development, and play a major role in division-site selection as well as nucleoid segregation in hyphae. As the various control systems that act to ensure that division occurs at mid-cell in rod-shaped bacteria are missing in Streptomyces sp., it was until now not clear how and where crosswall formation is initiated and completed in the multinucleoid hyphae. Our cryo-clet work sheds light on this process. A better understanding of vegetative cell division can lead to new insight into mycelial differentiation and morphogenesis within liquid grown samples, as well as fragmentation, and can lead to new strain improvement strategies for production purposes. Our study also illustrates the power of cryo-electron tomography and correlative techniques for elucidating the ultrastructural details of cell-cycle processes. Excitingly, we are only at the tip of the iceberg in terms of what can be discovered with these technologies. Improvement in cryolight resolution, as well as fluorescence signal enhancement of reporters (such as egfp) under cryo-conditions should achieve a long-term goal, namely the localization of protein complexes directly in high resolution images, and on native samples. Development of live-fluorescent imaging - prior to plunge freezing of samples for tomography - will make possible the capture of dynamic events as they are taking place. 20 20 ORIGIN # 3 jaargang 9, april 2014 Uit den ouden Doo Uit den ouden Doo Wie vaak door de Breestraat loopt zal het bordje misschien wel eens gezien hebben: In dit huis beheerste één man meer talen dan wie ook in Europa, vertelt het plakkaat bij nummer 113 ons. In dit pand woonde eind zestiende eeuw Josephus Justus Scaliger, toen hoogleraar aan de Universiteit Leiden. Hij bezat een uitgebreide kennis van onder andere het Latijn, Grieks, Hebreeuws, Aramees, Arabisch, Koptisch en Ethiopisch - een aantal van deze talen heeft hij zichzelf geleerd met behulp van oude geschriften. Vóór Scaliger werd aan de universiteit alleen de geschiedenis van het oude Griekenland en Romeinse Rijk bestudeerd; Scaliger was ook geïnteresseerd in andere oude beschavingen. Onder Scaligers hoogleraarschap werden ook de Joodse, Egyptische, Babylonische en Perzische geschiedenis toegevoegd aan het curriculum. In die periode stak de universiteit Leiden, dankzij Scaliger, met kop en schouders boven andere universiteiten in Europa uit op het gebied van onderwijs in klassieke en oosterse talen. Scaliger gaf geen college, maar leidde wel werkgroepen die bestonden uit geselecteerde excellente studenten. Hij hield zich zelf voornamelijk bezig met Bijbelonderzoek en was erg kritisch naar teksten van collega s, waarmee hij zich niet erg geliefd maakte. Toch bleef hij dankzij zijn kennis en geschreven werken in hoog aanzien staan in Leiden. Talen en geschiedenis waren de belangrijkste vakgebieden voor zijn Bijbelonderzoek. Volgens hem was slechte grammatica de enige oorzaak van onenigheid in religieuze geschriften. Een bekende uitspraak van hem is dan ook: "Utinam essem bonus grammaticus. Non aliunde discordiae in religione pendent quam ab ignoratione grammaticae" (Ik wil een goede grammaticus zijn. Religieuze verdeeldheid is enkel afhankelijk van onwetendheid over grammatica).
