2 1. The figure below shows pressure-volume loops and aortic flow signals recorded in a mouse heart during vena cava constriction (Georgakopoulos et al, AJP 1998). We model the systemic circulation of the mouse with a lumped parameter model that includes a time-varying elastance model of the ventricle, a constant-pressure preload, and a three-element windkessel afterload. a. Draw the model. b. Give the constitutive equation for the left ventricle. Give an estimate of the ventricular model parameters. c. Give the constitutive equations for the elements in the afterload model. Estimate the parameter values using the figure, or motivate what additional information is needed to make the estimation. d. Derive the differential equation the governs the time course of the arterial pressure during the filling phase of the ventricle.

3 2. To model the oxygen content c of blood (in ml O 2 /ml blood), we relate it to the partial pressure of oxygen po 2 according to the figure below: concentration [ml O 2 /ml blood] po [mmhg] 2 a. What is the name of this curve? What assumption is made by representing oxygen content exclusively through the relation in the figure? How valid is this assumption? We model the distribution of oxygen in the cardiovascular system with a lumped model. Estimate Q M for this case; use the figure above and estimates of additional parameters you might need. e. Estimate the diffusion coefficient D lung. Discuss how D lung is related to the physical properties of the lungs.

Noem minstens twee centrale regelmechanismen die in werking zullen treden om een dergelijke daling van de gemiddelde arteriële bloeddruk tegen te gaan. Leg uit hoe en op welke termijn de betreffende regelmechanismen een stijging van de gemiddelde arteriële bloeddruk bewerkstelligen. Als gevolg van deze bloeding daalt de gemiddelde arteriële ingangsdruk van een willekeurig gezond stuk spierweefsel (bijvoorbeeld een kuitspier) van 105 mmhg naar80mmhg(ziefiguur). Benoem het regelmechanisme waarmee weefsels en organen hun eigen perfusie kunnen reguleren en beschrijf kort hoe het werkt.

5 d. Met welke factor kan de perfusie van een skeletspier tijdens maximale inspanning toenemen als gevolg van het regelmechanisme uit de voorgaande vraag? In een ander stuk spierweefsel, waarvan het actieve perfusieregelmechanisme is uitgeschakeld, wordt de vaatweerstand gemeten gedurende eenzelfde daling van de gemiddelde arteriële ingangsdruk. Zie onderstaande figuur. e. Beredeneer waarom de vaatweerstand van dit passieve spierweefsel toeneemt.

6 4. Omdat het linker ventrikel de grootste massa heeft, wordt in deze opgave het hele hart samen als een afgeknotte ellipsoïde beschouwd. Voor de elektrische activatie van dit hart nemen we aan dat er slechts twee spierlagen zijn van elk één cel dik. De activatie van de binnenlaag is gelijktijdig. Dit geldt ook voor de buitenlaag. Hieronder vind je het verloop van de actiepotentialen van de binnen- en buitenlaag. a. Benoem de fasen in een actiepotentiaal. b. Geef de fasen aan in de tekening. c. Reconstrueer in de onderste grafiek het ECG-signaal in afleiding II op basis van de gegeven actiepotentialen en de gegeven aannames (in tijd kwantitatief, in voltage kwalitatief).

Teken in onderstaande grafiek hoe de actiepotentiaal van de binnenlaag eruit gaat zien en leg dit uit. e. Teken het bijbehorende ECG-signaal in afleiding II en leg uit. Veronderstel nu dat zowel de binnen- als de buitenlaag een verlaagde werking van de delayed rectifier potassium channels hebben. f.
