Abstract: Voorgaand onderzoek leert ons dat een milde stressbehandeling in staat is om geïsoleerde plantencellen aan te zetten tot dedifferentiatie en somatische embryogenese. Dit gebeurt door een samenspel met het plantenhormoon auxine. In dit project wordt, door experimenten op hormoon- en stressgerelateerde mutanten van Arabidopsis, dit samenspel verder onderzocht. Moleculaire Plantenfysiologie en Biotechnologie Geert Potters Rol van autofagie in normale en atherosclerotische arteriën Abstract: Autofagie is een herstellend, levensondersteunend proces dat met diverse pathologische condities is geassocieerd. Hoewel sommige histologische studies hebben aangetoond dat autofagie in geavanceerde atherosclerotische plaques voorkomt, is de rol van autofagie in atherogenese, plaquestabiliteit en vasomotorfunctie grotendeels onbekend. In het onderhavige onderzoeksvoorstel willen we een gedetailleerd onderzoek van de rol van autofagie in atherosclerose en in normale vasculaire functie uitvoeren. Fysiofarmacologie (FAR) Guido De Meyer Wim Martinet Rol van angiogenese in de darm in het ontstaan van extraintestinale articulaire manifestaties bij inflammatoir darmlijden Abstract: Recente data tonen aan dat een verhoogde angiongene activiteit aanwezig is in de aangetaste darm van patiënten met inflammatoir darmlijden (IBD). Via immunologisch onderzoek op menselijk darmweefsel en dieren experimenteel onderzoek willen we de rol van deze angiogenese beter omschrijven. Verder willen we onderzoeken of intestinale angiogenese bijdraagt tot het ontstaan van articulaire manifestaties bij IBD. Vakgroep Inwendige ziekten Martine De Vos Rol van angiogenese en angiogenetische groeifactoren (VEGF, PLGF) in portale hypertensie en fibrosevorming in diermodellen van cirrose, steatose en portale hypertensie. Abstract: Doel Het doel van dit project is de rol van placental growth factor (PLGF) in het ontstaan van portale hypertensie en zijn complicaties, in het ontstaan van cirrose en steatose/nash verder te bestuderen. - We zullen ons focussen op de rol van PLGF in de vorming van angiogenese in het mesenterium, van collateralen (shunten) in de perifere, splanchnische en pulmonale circulatie, waarbij ook de initiërende factoren zoals hypoxie (HIF cascade) zullen worden bestudeerd. De klinische complicaties van cirrose zullen worden nagegaan wanneer cirrose geïnduceerd wordt in PLGF-/- muizen. Er zal worden onderzocht of deze klinische complicaties kunnen voorkomen worden (preventie studies) door het toedienen van anti-angiogenetische farmaca (anti-plgf); kan de vorming van collateralen worden geïnhibeerd (zie orale presentatie C. Van Steenkiste), de longfunctie worden beïnvloed, de vorming van ascites worden verhinderd en meest belangrijk of de overleving kan worden verbeterd. Gezien patiënten zich meestal reeds in een stadium van cirrose of duidelijke PHT in de kliniek aanmelden, zal worden gekeken of elk van deze klinische complicaties reversibel zijn onder behandeling met anti-angiogenetische agentia. - De relatie tussen HSC, die fibrose induceren bij chronische leverschade, en angiogenese zal verder worden bestudeerd. Het effect van PLGF op HSC wordt bekeken, maar ook omgekeerd, of de HSC PLGF produceren en zo angiogenese kan induceren. Het gebruik van PLGF-/- muizen, anti-plgf en recombinant PLGF zal ons helpen om de rol van PLGF in fibrose te bestuderen. - In het muismodel van steatose en NASH zal de expressie (proteine/ mrna) van VEGF en PLGF met hun respectievelijke receptoren worden bepaald op verschillende tijdstippen in de lever, long en peritoneum. Het effect van het ontstaan van steatose, steatohepatitis en fibrose op de microcirculatie van peritoneum en lever (intravitaal microscopie en histologie), op collateraal vorming, portale druk en hemodynamica zal worden onderzocht. In een tweede tijd zal de rol van anti-angiogenetische factoren (anti-vegf, anti-plgf) in het ontstaan van steatohepatitis/fibrose in de NASH muismodellen worden bestudeerd. Celbiologie en Histologie Fysiologie ALBERT GEERTS Hendrik REYNAERT Leonardus VAN GRUNSVEN Rol van angiogenese en angiogenetische groeifactoren (VEGF, PLGF) in portale hypertensie en fibrose vorming in diermodellen van cirrose, steatose en portale hypertensie Abstract: Modellen van portale hypertensie en cirrose worden gebruikt om de preventie en reversibiliteit van collateraalvorming (in perifeer, splanchnisch en pulmonaal gebied) na te gaan, door muizen te behandelen met anti-plgf en door gebruik te maken van PLGF knock-out muizen. De fibrose vorming en de rol van hepatic stellate cellen in de angiogenese setting zal worden bestudeerd. In muismodellen van NASH wordt de rol van angiogenese in fibrose- en collateraalvorming in de verschillende stadia van steatose bestudeerd. Vakgroep Inwendige ziekten Isabelle Colle

2 Rol van aminozuurmetabolisme en peroxisomale enzymes in ziekteresistentie. Abstract: Deze aanvraag betreft co-financiering bij het Onderzoeksproject FWOAL411 met als onderwerp "Rol van aminozuurmetabolisme en peroxisomale enzymes in ziekteresistentie" Aanwijzingen voor een verband tussen aminozuurmetabolisme en ziekteresistentie werd verkregen in onderzoek naar ziekteresistentie van de tomatenmutant sitiens uitgevoerd aan het Laboratorium voor Fytopathologie (UGent, Prof. M. Höfte) en het Departement Planten Systeem Biologie (VIB-UGent, Dr. F. Van Breusegem). De ABA deficiënte sitiens mutant, die geraakt is in de laatste stap in de ABA-biosyntheseweg, heeft verhoogde resistentie tegen de necrotrofe schimmel Botrytis cinerea en de biotrofe schimmel Oidium neolycopersici. Wild type en sitiens planten werden met elkaar vergeleken 0 en 8 u na bladinfectie met B. cinerea met behulp van de Tom1 tomatenarrays. Hieruit bleken 240 genen significant verschillend geïnduceerd te worden ten gevolge van B. cinerea infectie, waaronder een reeks genen betrokken bij aminozuurmetabolisme. Veel van deze genen worden ook in Arabidopsis thaliana geïnduceerd na infectie met B. cinerea, en sommige genen tevens bij verschillende vormen van abiotische stress. Het doel van dit project is na te gaan of deze genen rechtstreeks betrokken zijn bij afweer tegen biotische en abiotische stressfactoren in Arabidopsis thaliana. Toegepaste Biologische Wetenschappen GEERT ANGENON Rol van aminozuur metabolisme en peroxisomale enzymes in ziekteresistentie. Abstract: De tomatenmutant sitiens, die deficiënt is in productie van abscisinezuur, is resistent aan necrotrofe fungi zoals Botrytis cinerea (Audenaert et al., 2002, Plant Physiol. 128: ) en heeft verminderde gevoeligheid aan de biotrofe fungus Oidium neolycopersici (Achuo et al., 2006, Plant pathology 55: ). Via microarray analyse werden in tomaat genen geïdentificeerd die na infectie differentieel geïnduceerd worden in wild type and sitiens planten. Dit project heeft als doel experimenteel na te gaan of deze differentieel gereguleerde genen een rol spelen in de afweer tegen pathogenen bij tomaat en Arabidopsis thaliana. Het project is een samenwerking tussen het Laboratorium Fytopathologie (UGent), het departement Plant Systems Biology (VIB/UGent) en het Laboratorium Plantengenetica (). Plantengenetica Toegepaste Biologische Wetenschappen Sigrid BRAUC GEERT ANGENON Eric DE WAELE Rol van abscisinezuur in de gevoeligheid van tomaat voor breedspectrum pathogenen en arbusculaire mycorrhizae. Abstract: Abscisinezuur (ABA) is een plantenhormoon dat, naast ethyleen en jasmonaat, een belangrijke rol speelt in de interactie tussen de plant en pathogene micro-organismen. Uit voorafgaand onderzoek aan de RUG is gebleken dat ABA mutanten veel resistenter zijn tegen Botridis cinerea en Sclerotinia sclerotiorum dan WT tomaten, maar geen verschil in gevoeligheid vertonen voor de biotrofe pathogeen Oidium lycopersici. De hypothese die we in dit onderzoek wensen te onderzoeken is dat (bepaalde) breedspectrum pathogenen na infectie een daling van het ABA gehalte verhinderen door zelf ABA te produceren of ABA productie door de plant te stimuleren (of afbraak door de plant te verhinderen) waardoor afweerreacties van de plant worden onderdrukt. Dit zou kunnen verklaren waarom interacties met deze pathogenen doorgaans compatiebeler zijn. Bovendien willen wij bekijken of breedspectrum AM een analoog mechanisme gebruiken om plant-afweer te onderdrukken. Plantenbiochemie Plantengroei en -ontwikkeling Els Prinsen Rol van ABCC6, een transmembranaire transporter in de lever, in bindweefsel homeostase met Pseudoxanthoma Elasticum als studiemodel Abstract: Dit project beoogt de opheldering van de identiteit van het substraat van ABCC6, een transmembraan transporter die verstoord is door een erfelijke recessieve genodermatose, Pseudoxanthoma Elasticum (PXE). Tevens wordt beoogd het verband met een gestoorde calciumhomeostase die resulteert in abnormale bindweefselmineralisatie in deze aandoening op te helderen. Tenslotte zal ook de rol van vitamine K in deze aandoening geëploreerd worden. Vakgroep Pediatrie en genetica Anne De Paepe Rol van aandachtsmechanismen en cognitieve controle bij depressie: een experimenteel onderzoek naar de onderliggende neuro-biologische werkingsmechanismen Abstract: Recent neurocognitief onderzoek heeft stemmingsstoornissen gerelateerd aan een dificit in de strategische aandachtprocessen geleid door een frontocorticaal-subcorticaal netwerk. De doelstelling van dit project is om via rtms en ERP studies de functionele prefrontale aandachtsdefecten binnen dit netwerk te onderzoeken waardoor de relatie tussen cognitieve controle en klinische symptomatologie wordt onderzocht. Er wordt gebruik gemaakt van experimentele aandachtsparadigma's. 3 Abstract: Chronische musculoskeletale aandoeningen vormen een groeiend probleem met een grote impact op het functioneren en de levenskwaliteit van de patiënt, alsmede op de gezondheidszorg en economische systemen (afwezigheid op het werk, ziektekostenverzekering). Rehabilitatie kan de pijn doen dalen en het functioneel niveau van chronische pijn verbeteren. Echter, een gebrek aan motivatie en adherentie met oefenprogramma's belemmert het succes van een optimale behandelingsuitkomst, waarbij de patiënt geen gebruik maakt van zijn/haar volledig potentieel voor herstel. Het voorgestelde project ontwikkeld multi-perspectieve motivationele en overtuigende strategieën die benut kunnen worden om de patiënt warm te maken en te overhalen om alle mogelijke middelen te benutten om de uitkomst van de behandeling te optimaliseren. Drie aspecten staan centraal in dit onderzoek: 1) de definitie van een conceptueel model voor een multi-perspectieve strategie om motivatie te verbeteren en de patiënt over te halen tot optimale naleving van de behandeling gedurende musculoskeletale rehabilitatie, 2) de realisatie van technologie-ondersteunde overtuigende rehabilitatie die het ontwikkelde concept van motivationele en persuasieve strategieën voor rehabilitatie in musculoskeletale aandoeningen steunt, en 3) de definitie en toepassing van een multi-perspectieve evaluatie raamwerk voor rol-gebaseerde persuasieve rehabilitatie in musculoskeletale aandoeningen. Een focus van het project zal zijn om te patiënt te bereiken, voorbij de rehabilitatiesessies met beroepsbeoefenaren in de gezondheidszorg, in zijn/haar natuurlijke omgevingscontext thuis, op werk, of gedurende zijn/haar vrije tijd. Revalidatiewetenschappen Expertisecentrum voor Digitale Media Biomedisch Onderzoeks Instituut Karin CONINX Annick TIMMERMANS Rollen van Nanos familieleden in de progressie van kanker Abstract: Nanos-eiwitten binden op boodschapper-rna's, die normaliter verantwoordelijk zijn voor de synthese van eiwitten die essentieel zijn voor de correcte embryonale ontwikkeling van vooral de geslachtsorganen. Op die manier reguleren Nanos-eiwitten de stabiliteit van deze belangrijke RNA's. Wij ontdekten dat deze Nanos-eiwitten in bepaalde tumoren (bv. in de prostaat of long) op abnormaal hoge wijze aangemaakt worden. Bovendien bleek zulke activering de kwaadaardige karakteristieken van de tumorcellen te verhogen. Ons project beoogt deze fenomenen grondig te bestuderen, zodat deze kennis hopelijk kan leiden tot een verbeterde diagnose of mogelijk zelfs betere therapie van dergelijke tumoren. Onze studies omvatten meerdere hoog-technologische invalswegen zodat de kansen dat nieuwe en bruikbare inzichten worden verworven zo groot mogelijk zijn. Abstract: (Niet beschikbaar in het Nederlands) The aim of my research is to study the establishment of the body axes and the early patterning of vertebrate embryos. The main focus will be the understanding of mouse embryonic development but using the guiding principle that this is a common process to all vertebrates. Thus while the mouse will be the model organism I will focused on, I will also use the frog as a 'test tube' as it is an more manipulable organism. This double approach allows a quick testing of an hypothesis in the frog before moving to heavier experiments that will be carried on in the mouse. In the long term, my goal is to identify and characterize genes that are involved in the inductive and determinative processes that occur during the gastrulation, neurulation and segmentation stages of development. These genes will most likely be signaling and regulatory factors. For the present, I will start by studying in more details the signaling factor, Wnt-8 and its antagonist, Frzb-1, both secreted proteins. Wnt-8 is one of the members of a large family of homologous genes found in organisms as diverse as the worm, the fly and the vertebrates. Moreover, some of these genes have been originally isolated in mouse because of their capacity to induce breast cancer. During my postdoctoral research, I discovered in mouse and frog a new factor, Frzb-1 that behaves as a secreted antagonist of Wnt-8. More genes of the same family were subsequently discovered but none of them seems to be involved in the gastrulation process. It has been shown in the frog that the Wnt-8 factor is involved in the induction of mesoderm during the gastrulation process and its patterning, leading to the formation of muscle tissue. Until now all the functional studies have been done in the frog with DNA and RNA injection during the first cleavages of the embryo. This leads after translation and traduction to immediate secretion of the proteins in the extracellular space in the descendants of the injected cell(s). In contrast, in the embryo the Wnt-8 and Frzb-1 genes start normally to be expressed at a later stage, the gastrulation. The role of these factors can thus not be studied in a controlled temporal fashion similar to the normal situation nor in a restricted population of cells. The way I propose to circumvent these difficulties is to study the differentiation events in embryo explants that are placed in presence of conditioned medium containing the Wnt-8 and/or Frzb-1 proteins. The major obstacle to this approach is that although the first Wnt gene was discovered in 1982, no protein of any members of the vertebrate Wnt family has

4 been shown to be secreted in detectable amount in expression systems, precluding any direct study of the Wnt proteins. Recently, I decide to tackle this problem and found that by transfecting the frog Wnt-8 gene carrying an epitope tag for an easy detection of the protein in a human embryonic kidney cell line, I could obtain substantial amount of the protein in a soluble form in the conditioned medium of these cells. This is the first case where a vertebrate Wnt protein is obtained in significant amount. Thanks to this breakthrough and the availability of secreted Frzb-1 protein, produced by a similar technique, I will study the effects of the conditioned medium containing the Wnt-8 protein, alone or with the addition of its antagonist, Frzb-1, on the determination and differentiation of embryo explants. I will first use frog embryo explants to test the proposed role of Wnt-8 in mesoderm patterning and muscle formation. For that purpose, I will dissect small regions of the embryo prior to or at the beginning of gastrulation. These explants will correspond either to the future ventral mesoderm that normally differentiate in blood or to the prospective neuroectodermal region that would form only neurons and epidermis. The incubation of these explants in presence of Wnt-8 will permit to test the hypothesis as the neuroectoderm may be induced to form mesoderm and the ventral mesoderm may differentiate in muscle. To analyze these events I will use two techniques, first a histological analysis of explants cultivated for several days to observe the various cell types that differentiate and second, a reverse-transcription PCR assay to detect the induction of determining genes a few hours after the beginning of the incubation of the explants. Subsequently, I plan to develop the technique of mouse embryo explants to continue analyzing the role of Wnt-8 factor in this model organism. This approach is entirely new and should open new ways of studying the early mouse development by allowing to analyze the role of different factors alone and in combination on the determination and differentiation processes. This would help finding which are the signaling molecules that are involved in mouse embryogenesis. Uitgaande van proefdieremodellen zal het oorzakelijk verband tussen M. tuberculosis vermenigvuldiging, versoorde macrofaag cholesterol metabole reactiewegen en de cholesterolaard van mycolzuren. Vakgroep Biomedische Moleculaire Biologie Johan Grooten Role of the Hxt5 glucose transceptor in the concerted yeast nutrient sensing. Abstract: Until now, it has been assumed that the glucose requirement for activation of the FGM pathway was fullfilled by glucose sensing via the Ras/cAMP pathway. Recently, Hxt5, a hexose transporter, has been found to have a exceptional expression pattern. Like the FGM transceptors, Hxt5 is upregulated during conditions in which yeast growth is not sustained. Combined with the extraodinary long cytosolic N-terminal domain of Hxt5 this finding led to the hypothesis that Hxt5 might act as a glucose transceptor for the FGM pahtway. Preliminary data revealed that activation of thefgm pathway indeed relied on the presence of Hxt5. Patrick Van Dijck Giovanni Rodriguez Leguizamon Role of the camp-pka pathway on adhesion and pathogenicity in the humanfungal pathogen Candida glabrata. Abstract: Candida glabrata is an upcoming human fungal pathogen, frequently resistant to azole antifungal drugs and therefore difficult to treat clinically. One of the first steps in the infection process is adhesion to a surface, in which particular cell wall proteins play an important role. C. glabrata has a large group of such adhesins called the EPA-family. It isalready known that glucose has an effect on the expression of EPA1-3 via the putative receptor Gpr1, a homologue of S. cerevisiae Gpr1. The PKApathway, which is important for virulence in many fungal pathogens (eg.c. albicans), is a strong candidate for connecting Gpr1 to EPA gene expression. In this project, Gpr1 will be first investigated further in order to prove it is a true sensor for glucose, sucrose and fructose. Therefore we will make specific point mutants to abolish signaling and search for competitive inhibitors of signaling. 5 Abstract: Fungal airway diseases are chronic eosinophilic mucosal inflammatory disorders driven by hypersensitivity responses to extramucosal fungi extracts1. Staphylococcus aureus, a prodigious superantigen producer, is commonly cultured from fungal sinus surgeries1,2. Although the role of superantigens in chronic rhinosinusitis has recently been recognized3,4, similar immunological mechanisms in fungal airway disease need to be explored1,5-7. Superantigens as a cofactor in fungal disease immunopathogenesis will open new horizons in treatment.
